The shapes DT is using are the standard open tuning chord shapes, but with a first finger barre. So for example at the 5th fret in open E the barre is A, then when you make the 4 chord shape with fingers 2 and 3, you get a D chord....etc. It is kind of copying how a pianist would play shuffles and maybe originates from that, Jerry Lee Lewis and Fats Domino. Among others, this is used in many Rolling Stones songs, but mostly in open G. I have recorded a few songs using those chords shapes, one that springs to mid is "Love" on the Second Mind album, which was supposed to sound like it kind of fell off the back of Exile On Main Street.
Mrs T is in standard tuning and playing chords that you can find in any guitar instruction manual. You need to spend some time learning the structure of the song they are playing. What they are doing reminds me of Delaney & Bonnie. It is really important to understand the structure before you try to decode the various improvisations.

At about 3:05 it looks to me like he slides a barred G up to A and then hits open strings and appears to hit the low E as well.
